The meaning behind the asterisk in the Thunderbolts* title has finally been explained: The New Avengers. Promotional materials for the movie, including social media accounts, ads, theater standees, and more, have been changed to reflect the new title after opening weekend.
However, according to Deadline, Disney is not technically re-naming Thunderbolts*, it's just spelling out that asterisk/subtitle. It all makes sense after the final scene in the movie, where Yelena Belova (Florence Pugh), Bucky Barnes (Sebastian Stan), Red Guardian (David Harbour), Ghost (Hannah John-Kamen), and John Walker (Wyatt Russell) are given the name of The New Avengers by Valentina Allegra de Fontaine (Julia Louis-Dreyfus).
The studio even had longtime Marvel Cinematic Universe cast member Stan making updates to his character poster in person.

The story is one of the more meaningful ones Marvel has done, tackling mental health and depression through its main group of characters.
"If you are going to tell a story about something like this, the last thing we want for anyone that struggles, including myself, with what the characters are going through is to feel like we’re being reductive about it or that we’re simplifying it or that we’re saying that it can be even solved," Director Jake Schreier expressed to The Hollywood Reporter. "It’s more about the idea that you can bear it with others, but it’s not going to go away."
Pugh, Stan, Harbour, John-Kamen, and Russell are all confirmed to return for Avengers: Doomsday, which was announced prior to the film's release.
"I was aware of it at the same time the rest of the world was. It’s hard with these movies. There’s a lot of attention on them, there’s a lot of marketing, and there’s a lot of stuff that gets out there," he commented. "So there are things that might have, in some contexts, remained uncertain for people who are really paying attention going into [Thunderbolts*]. But it’s nice to feel like this is all building to something and that it’s part of a larger world and context. When I would worry about spoilers or things like that, Jon [Watts] would always say, 'When you sit down in the theater and the lights go down, you forget all of that stuff. You’re in the story.' And maybe that’s something that Kevin told Jon."

Screenwriter Eric Pearson told ScreenRant, "The New Avengers thing was Kevin's idea when I first brought them this project. It was really just the skeleton of the movie that you see, and when I got to the end, I always had that moment with Yelena right behind Valentina, saying, 'You work for us now.' Valentina was being forced to introduce the Thunderbolts."
She continued to explain, "I've done a lot of pitching at Marvel, and you get a lot of notes. But what all Kevin said was, 'I think that they should be the New Avengers. I think she should announce them as the New Avengers.' I thought, 'Man, if that's the only note I'm getting from this whole thing? Take the money and run. Get out of here. This is great!' I didn't know what the plan was for that, but that's not my job. I'm writing this movie, and I want to write the Yelena, Bucky, Bob, Val, and Walker story. I want to write that story. So, that's another guy's problem down the line, in terms of what it means for them to be the New Avengers."
