Chark suffered the injury on the third play of Thursday night's game. He was blocking when running back James Robinson accidentally rolled into his left leg while being tackled by linebacker Germaine Pratt.
The 25-year-old has caught seven passes for 154 yards and two touchdowns in three games this season.
It's unclear if Chark will be out for the remainder of the season, but "out indefinitely" was certainly not the news Jaguars fans were hoping for after an 0-4 start.
Chark is in his fourth season with the Jaguars, who selected him in the second round of the 2018 draft. His best season came in 2019, when he caught 73 passes for 1,008 yards and eight touchdowns.
The LSU product is in the final year of his rookie deal, and it remains to be seen how his ankle injury will impact his next contract.
