With a bevy of roster moves this week, it is probably best to look at them in a few different articles to give them the proper amount of analysis.
First, let’s take offense.
The Colts this week have sent OG Seth Olsen to Injured Reserve (with the “Designated to Return” label) and waived TE/FB Dominique Jones. Two players who have seen significant snaps and both started games for the Colts this season.
So, as we can see, it was all subtraction of personnel on the offensive side of the ball but let’s take a look at possible gameday adjustments that are likely to occur to help compensate.
With Olsen going to IR-DTR, he should be able to return to the active roster, provided he’s healthy, after 8 weeks on the shelf. The Colts apparently value his experience and ability to use this designation for him as they can only use this new roster management tool with 1 player per season.

Colts rookie running back Vick Ballard will make his first NFL start Sunday against the New York Jets after Donald Brown had surgery to repair torn cartilage in his knee.
Interim coach Bruce Arians says Brown had surgery Tuesday and shouldn't miss more than two to three weeks.

According to Mike Chappell of the Indy Star, Colts outside linebacker Robert Mathis will be out for the next 23 weeks with a sprained left knee.This is a big blow to the Colts’ defense. They paid Mathis big money to re-sign in Indy while they re-tool as an organization.
Andrew Luck had his best game yet in leading the Colts back from a 21-3 halftime deficit over the Green Bay Packers. It ties the largest comeback win of the season, and the largest by a rookie since Matthew Stafford’s 21-point comeback over Cleveland in 2009.

Indianapolis Colts running back Donald Brown underwent a knee scope and will miss two to three weeks. The injury occurred late in the second half against the Packers.
Rookie tailback Vick Ballard will get the start against the Jets on Sunday. Ballard is a nice sleeper pick in all formats as the Jets have been terrible against the run this season.
